Transcriptomic Approaches To Identify Receptors And Downstream Targets

We are using micro-dissection to recover individual sensory neurons required for sensing ascarosides in C. elegans. We will use next-generation sequencing approaches such as RNAseq to identify their transcript profiles from these isolated cells. To characterize both general and sex-specific effects of these chemicals, we will profile neurons from both the sexes to identify sex-specific receptors and other molecules. Candidates from this data set will be tested using RNAi in a bioassay for response to the individual chemicals or mixtures in a genetic background sensitized to neuronal RNAi.
